    Pharmacokinetics of chloroquine in rats

Chloroquine has been extensively used in mass drug administrations, which may have contributed to the emergence and spread of resistance. It is recommended to check if chloroquine is still effective in the region prior to using it.

Abstract. Effect of Gnetum africana on the pharmacokinetic parameters of chloroquine was investigated. Chloroquine phosphate 200 mg/Kg was concurrently administered to overnight fasted albino rats. Blood samples were collected 15, 30, 60, 120, 240 and 480 minutes after administration through cardiac puncture. The pharmacokinetics of chloroquine do not differ to a clinically important extent between black and white patients 11, 69, 79. The average melanin content of a black person is estimated to be 1 g and for a white person is estimated to be 250 mg. The Centers for Disease Control and Prevention recommend against treatment of malaria with chloroquine alone due to more effective combinations. In areas where resistance is present, other antimalarials, such as mefloquine or atovaquone, may be used instead.

  3. The single oral dose pharmacokinetics of chloroquine was studied alone and after coadministration with phytomedicines NIPRID/92/001/1-1 AM-1, Niprisan®, and Nifadin® in rats. Plasma chloroquine concentrations were measured using High performance liquid chromatography HPLC method developed earlier in our laboratory. The data were fitted into a WinNonlin standard non-compartmental.
