    Chloroquine mechanism of action autophagy

Chloroquine has been extensively used in mass drug administrations, which may have contributed to the emergence and spread of resistance. It is recommended to check if chloroquine is still effective in the region prior to using it.

MHY1485 has the ability to penetrate the cell. It prevents the ultraviolet-induced oxidative stress in keratinocytes and fibroblasts. MHY1485 is mTOR activator that potently inhibits autophagy by suppression of fusion between autophagosomes and lysosomes. The autophagy inhibitors, chloroquine and bafilomycin A1, as well as genetic suppression of the autophagic signaling proteins Atg5 or Atg 7 attenuated the impact of the combination treatment of 1. In parallel with these findings, another study demonstrated that autophagy is a key mechanism for anti-leukemic effects of ATO in AML cell lines and leukemic progenitors from AML patients, and this induction of autophagy flux is associated with mitogen-activated protein kinase kinase MEK/extracellular signal-regulated kinase ERK-dependent autophagic cell death.

The Centers for Disease Control and Prevention recommend against treatment of malaria with chloroquine alone due to more effective combinations. In areas where resistance is present, other antimalarials, such as mefloquine or atovaquone, may be used instead.

  4. To investigate the impact of autophagy in either promoting or protecting CLL cells from death, we used chloroquine, an agent that prevents fusion of the autophagosome with the lysosome. 30 In these experiments, chloroquine did indeed inhibit autophagosome-lysosome fusion at 0.1-0.5μM, concentrations attainable in patients receiving this for malaria treatment 31 supplemental Figure 2.

Inhibition of autophagy see section on mechanisms of action, measured by counting the number of autophagic vacuoles per cell in tumour tissues and peripheral blood mononuclear cells of patients, was only noted in patients receiving at least 1200 mg HCQ daily. This study recommends an adjuvant HCQ dose of 600 mg, twice daily. Chloroquine is a 9-aminoquinoline that has been known since 1934. Specifically synthesised to be used as an antimalarial agent, chloroquine was subsequently shown to have immunomodulatory properties that have encouraged its application in the treatment of autoimmune diseases such as rheumatoid arthritis. Oct 01, 2018 Mechanism of Action Chloroquine, a 4-aminoquinoline, is an anti-protozoal agent. The precise mechanism by which Chloroquine exhibits activity is not known. Chloroquine, may exert its effect against Plasmodium species by concentrating in the acid vesicles of the parasite and by inhibiting polymerization of heme. It can also inhibit certain enzymes by its interaction with DNA.
